    Opposing effects of dopamine antagonism in a motor sequence task—tiapride increases cortical excitability and impairs motor learning

    The dopaminergic system is involved in learning and participates in the modulation of cortical excitability (CE). CE has been suggested as a marker of learning and use-dependent plasticity. However, results from separate studies on either motor CE or motor learning challenge this notion, suggesting opposing effects of dopaminergic modulation upon these parameters: while agonists decrease and antagonists increase CE, motor learning is enhanced by agonists and disturbed by antagonists. To examine whether this discrepancy persists when complex motor learning and motor CE are measured in the same experimental setup, we investigated the effects of dopaminergic (DA) antagonism upon both parameters and upon task-associated brain activation. Our results demonstrate that DA-antagonism has opposing effects upon motor CE and motor sequence learning. Tiapride did not alter baseline CE, but increased CE post training of a complex motor sequence while simultaneously impairing motor learning. Moreover, tiapride reduced activation in several brain regions associated with motor sequence performance, i.e. dorsolateral PFC, supplementary motor area, Broca's area, cingulate and caudate body. Blood-oxygenation-level-dependent ( BOLD) intensity in anterior cingulate and caudate body, but not CE, correlated with performance across groups. In summary, our results do not support a concept of CE as a general marker of motor learning, since they demonstrate that a straightforward relation of increased CE and higher learning success does not apply to all instances of motor learning. At least for complex motor tasks that recruit a network of brain regions outside motor cortex, CE in primary motor cortex is probably no central determinant for learning success

    A Map of Periodicity Orthogonal to Frequency Representation in the Cat Auditory Cortex

    Harmonic sounds, such as voiced speech sounds and many animal communication signals, are characterized by a pitch related to the periodicity of their envelopes. While frequency information is extracted by mechanical filtering of the cochlea, periodicity information is analyzed by temporal filter mechanisms in the brainstem. In the mammalian auditory midbrain envelope periodicity is represented in maps orthogonal to the representation of sound frequency. However, how periodicity is represented across the cortical surface of primary auditory cortex (AI) remains controversial. Using optical recording of intrinsic signals, we here demonstrate that a periodicity map exists in primary AI of the cat. While pure tone stimulation confirmed the well-known frequency gradient along the rostro-caudal axis of AI, stimulation with harmonic sounds revealed segregated bands of activation, indicating spatially localized preferences to specific periodicities along a dorso-ventral axis, nearly orthogonal to the tonotopic gradient. Analysis of the response locations revealed an average gradient of − 100° ± 10° for the periodotopic, and −12° ± 18° for the tonotopic map resulting in a mean angle difference of 88°. The gradients were 0.65 ± 0.08 mm/octave for periodotopy and 1.07 ± 0.16 mm/octave for tonotopy indicating that more cortical territory is devoted to the representation of an octave along the tonotopic than along the periodotopic gradient. Our results suggest that the fundamental importance of pitch, as evident in human perception, is also reflected in the layout of cortical maps and that the orthogonal spatial organization of frequency and periodicity might be a more general cortical organization principle

    Differential effects of synchronous and asynchronous multifinger coactivation on human tactile performance

    <p>Abstract</p> <p>Background</p> <p>Repeated execution of a tactile task enhances task performance. In the present study we sought to improve tactile performance with unattended activation-based learning processes (i.e., focused stimulation of dermal receptors evoking neural coactivation (CA)). Previous studies show that the application of CA to a single finger reduced the stationary two-point discrimination threshold and significantly increased tactile acuity. These changes were accompanied by an expansion of the cortical finger representation in primary somatosensory cortex (SI). Here we investigated the effect of different types of multifinger CA on the tactile performance of each finger of the right hand.</p> <p>Results</p> <p>Synchronous and asynchronous CA was applied to all fingers of a subject's dominant hand. We evaluated changes in absolute touch thresholds, static two-point discrimination thresholds, and mislocalization of tactile stimuli to the fingertips. After synchronous CA, tactile acuity improved (i.e., discrimination thresholds decreased) and the frequency of mislocalization of tactile stimuli changed from directly neighboring fingers to more distant fingers. On the other hand, asynchronous CA did not significant improve tactile acuity. In fact, there was evidence of impaired tactile acuity. Multifinger CA with synchronous or asynchronous stimulation did not significantly alter absolute touch thresholds.</p> <p>Conclusion</p> <p>Our results demonstrate that it is possible to extend tactile CA to all fingers of a hand. The observed changes in mislocalization of tactile stimuli after synchronous CA indicate changes in the topography of the cortical hand representation. Although single-finger CA has been shown to improve tactile acuity, asynchronous CA of all fingers of the hand had the opposite effect, suggesting the need for synchrony in multifinger CA for improving tactile acuity.</p

    Repetitive Electric Stimulation Elicits Enduring Improvement of Sensorimotor Performance in Seniors

    Age-related changes occur on all stages of the human somatosensory pathway, thereby deteriorating tactile, haptic, and sensorimotor performance. However, recent studies show that age-related changes are not irreversible but treatable through peripheral stimulation paradigms based on neuroplasticity mechanisms. We here applied repetitive electric stimulation (rES) to the fingers on a bi-weekly basis for 4 weeks to induce enduring amelioration of age-related changes in healthy individuals aged 60–85 years. Tactile, haptic, and motor performance gradually improved over time of intervention. After termination of rES, tactile acuity recovered to baseline within 2 weeks, while the gains in haptic and motor performance were preserved for 2 weeks. Sham stimulation showed no comparable changes. Our data indicate that age-related decline of sensorimotor performance can be ameliorated by rES and can be stabilized by the repeated application. Thus, long-term application of rES appears as a prime candidate for maintaining sensorimotor functions in elderly individuals

    Improvement of sensorimotor functions in old age by passive sensory stimulation

    Sensorimotor functions decrease in old age. The well-documented loss of tactile acuity in elderly is accompanied by deterioration of haptic performance and fine manipulative movements. Physical training and exercise can maintain sensorimotor fitness into high age. However, regular schedules of training require discipline and physical fitness. We here present an alternative interventional paradigm to enhance tactile, haptic, and fine motor performance based on passive, sensory stimulation by means of tactile coactivation. This approach is based on patterned, synchronous tactile stimulation applied to the fingertips for 3 hours. The stimulation drives plastic reorganizational changes in somatosensory cortex that affect perception and behavior: We demonstrate that following 3 hours of coactivation tactile acuity as well as haptic object exploration and fine motor performance are improved for at least 96 hours. Because this kind of intervention does not require active participation or attention of the subjects, we anticipate that coactivation is a prime candidate for future therapeutic interventions in patients with impaired sensorimotor abilities. It can be assumed that the maintenance and restoration of sensorimotor functions can ensure and preserve independence of daily living. Further optimizing of the stimulation protocol can be assumed to strengthen both the range and durability of its efficacy

    Bodily training typically evokes behavioral and perceptual gains, enforcing neuroplastic processes and affecting neural representations. We investigated the effect on somatosensory perception of a three-day Zen meditation exercise, a purely mental intervention. Tactile spatial discrimination of the right index finger was persistently improved by only 6 hours of mental-sensory focusing on this finger, suggesting that intrinsic brain activity created by mental states can alter perception and behavior similarly to external stimulation

    Superior Sensory, Motor, and Cognitive Performance in Elderly Individuals with Multi-Year Dancing Activities

    Aging is associated with a progressive decline of mental and physical abilities. Considering the current demographic changes in many civilizations there is an urgent need for measures permitting an independent lifestyle into old age. The critical role of physical exercise in mediating and maintaining physical and mental fitness is well-acknowledged. Dance, in addition to physical activity, combines emotions, social interaction, sensory stimulation, motor coordination and music, thereby creating enriched environmental conditions for human individuals. Here we demonstrate the impact of multi-year (average 16.5 years) amateur dancing (AD) in a group of elderly subjects (aged 65–84 years) as compared to education-, gender- and aged-matched controls (CG) having no record of dancing or sporting activities. Besides posture and balance parameters, we tested reaction times, motor behavior, tactile and cognitive performance. In each of the different domains investigated, the AD group had a superior performance as compared to the non-dancer CG group. Analysis of individual performance revealed that the best participants of the AD group were not better than individuals of the CG group. Instead, the AD group lacked individuals showing poor performance, which was frequently observed for the CG group. This observation implies that maintaining a regular schedule of dancing into old age can preserve cognitive, motor and perceptual abilities and prevent them from degradation. We conclude that the far-reaching beneficial effects found in the AD group make dance, beyond its ability to facilitate balance and posture, a prime candidate for the preservation of everyday life competence of elderly individuals

    Cortical excitability in human somatosensory and visual cortex: implications for plasticity and learning – a minireview

    The balance of excitation and inhibition plays a key role in plasticity and learning. A frequently used, reliable approach to assess intracortical inhibition relies on measuring paired-pulse behavior. Moreover, recent developments of magnetic resonance spectroscopy allows measuring GABA and glutamate concentrations. We give an overview about approaches employed to obtain information about excitatory states in human participants and discuss their putative relation. We summarize paired-pulse techniques and basic findings characterizing paired-pulse suppression in somatosensory (SI) and (VI) visual areas. Paired-pulse suppression describes the effect of paired sensory stimulation at short interstimulus intervals where the cortical response to the second stimulus is significantly suppressed. Simultaneous assessments of paired-pulse suppression in SI and VI indicated that cortical excitability is not a global phenomenon, but instead reflects the properties of local sensory processing. We review studies using non-invasive brain stimulation and perceptual learning experiments that assessed both perceptual changes and accompanying changes of cortical excitability in parallel. Independent of the nature of the excitation/inhibition marker used these data imply a close relationship between altered excitability and altered performance. These results suggest a framework where increased or decreased excitability is linked with improved or impaired perceptual performance. Recent findings have expanded the potential role of cortical excitability by demonstrating that inhibition markers such as GABA concentrations, paired-pulse suppression or alpha power predict to a substantial degree subsequent perceptual learning outcome. This opens the door for a targeted intervention where subsequent plasticity and learning processes are enhanced by altering prior baseline states of excitability

    Rapid Assessment of Age-Related Differences in Standing Balance

    As life expectancy continues to rise, in the future there will be an increasing number of older people prone to falling. Accordingly, there is an urgent need for comprehensive testing of older individuals to collect data and to identify possible risk factors for falling. Here we use a low-cost force platform to rapidly assess deficits in balance under various conditions. We tested 21 healthy older adults and 24 young adults during static stance, unidirectional and rotational displacement of their centre of pressure (COP). We found an age-related increase in postural sway during quiet standing and a reduction of maximal COP displacement in unidirectional and rotational displacement tests. Our data show that even low-cost computerized assessment tools allow for the comprehensive testing of balance performance in older subjects

    Touch improvement at the hand transfers to the face

    SummaryThe hand–face border is one of the most prominent features of the primate somatosensory cortex. A reduction of somatosensory input, following amputation or anesthesia, induces perceptual changes across this border that are explained by plastic competitive mechanisms [1–4]. Whether cross-border plasticity can be induced by learning processes relying on increased somatosensory input has been unclear. Here we report that training-independent learning [5] improves tactile perception, not only at the stimulated index finger, but also at the unstimulated face. These findings demonstrate that learning-induced tactile improvement can cross the hand–face border, suggesting that facilitation-based plasticity may operate in the healthy human brain
